Makes a huge difference, a running back gets no head of steam when he receives the ball in the shotgun. There is a bigtime difference. Even Brian Westbrook said thisd last week when asked about it.

Sure - but that still assumes that the O-line is actually blocking the defense. Otherwise it will be the D-lineman that takes the hand-off.
